What is/are Element 1?

The Repressor Element 1 Silencing Transcription Factor (REST) also known as Neuron-Restrictive Silencing Factor (NRSF) has been reported as a neuroprotective molecule in certain neurological disorders. ^[1] The expression of most of the genes required for axon growth and guidance and for synapsis formation is repressed by a single master transcriptional regulator, the Repressor Element 1 Silencing Transcription factor (REST). ^[2] BACKGROUND Repressor element 1-silencing transcription factor (REST) is a master regulator that is highly expressed in multipotent stem cells to repress gene networks involving a wide range of biological processes. ^[3] Repressor element 1-silencing transcription factor (REST) plays a crucial role in the differentiation of neural progenitor cells (NPCs). ^[4] A subset of breast cancers express normal levels of REST (repressor element 1 silencing transcription factor) mRNA but lack functional REST protein. ^[5] The transcriptional repressor REST (Repressor Element 1-Silencing Transcription factor) is a focus of interest as it is consistently upregulated in epilepsy patients and following brain insult in animal models of epilepsy and ischemia. ^[6] We show that the developmental deficits of interneurons may be linked to increased levels of the Repressor Element 1-Silencing Transcription factor (REST), which we report to bind to Gad1, which encodes glutamate decarboxylase (GAD) 67, a rate-limiting enzyme in the production of GABA. ^[7] REST (RE1 silencing transcription factor, also called NRSF) is a zinc finger transcription factor that binds to the conserved motif repressor element 1 found in the promoter regions of many neuronal genes with functions in neuronal development and differentiation. ^[8] The repressor element 1-silencing transcription factor/neuron-restrictive silencer factor (REST/NRSF) binds to repressor element 1/neuron-restrictive silencer element (RE1/NRSE) sites in the genome and recruits effector proteins to repress its target genes. ^[9] Transcriptional suppressor, repressor element 1-silencing transcription factor (REST, neuron-restrictive silencing factor), is necessary and sufficient for the development of chronic hyperalgesia after nerve injury or inflammation. ^[10] Recent studies show that levels of repressor element 1-silencing transcription factor (REST), increases with normal aging in mice and humans, and reduces neuronal excitation. ^[11]
