Temporally Programmed 一時的にプログラムされた - Until now, synthetic efforts have majorly contributed to fundamental strategies; however, temporally programmed functions are rarely explored. [1] To address this dilemma, genetic engineering of a body temperature‐responsive and matrix metalloproteinase (MMP)‐cleavable conjugate of interferon alpha (IFNα) and elastin‐like polypeptide (ELP) is reported with spatiotemporally programmed two‐step release kinetics for tumor therapy. [2] An efficacious temporally programmed combination therapy of extended half-life interleukin 2 (eIL2), tumor targeting antibody, and interferon (IFN) α was recently reported; however, tumor-ablative efficacy was associated with significant toxicity. [3] We also discuss key challenges and opportunities for translation of temporally programmed treatment schedules to the clinic, including alignment of immunological timescales in preclinical models and humans, and the use of current and emerging biomarkers.
