3d Coculture - We report that high‐dose taPIT spares 25% of the local immune cell population, five times more than the conventional PDT regimen, in a 3D coculture model incorporating epithelial ovarian cancer cells and T cells. ^[1] We found that epithelial polarity loss also occurs in a 3D coculture system that combines acini with human mammary adipose tissue, and establish that a paracrine effect of the tissue adipokine leptin causes loss of polarity by overactivation of the PI3K/Akt pathway. ^[2] The promoting effect was also demonstrated in a 3D coculture model. ^[3] In this study, primary human MCs were generated from CD34+ progenitor cells and a 3D coculture model was developed to study the interplay between colon cancer cells and MCs. ^[4] The 3D coculture model appeared to successfully generate prevascularized constructs and were sufficient in exceeding the ~200 μm diffusion limit. ^[5] The fluorescence kinetics study showed that both EGFP and Ds-red signals in the 3D coculture could be simultaneously detected without interference with each other. ^[6] The 3D coculture was used to evaluate cytotoxicity or IC50 of three antitumor drugs: tamoxifen, oxaliplatin and cisplatin. ^[7] This study proposed a conceptual 3D coculture model based on collagen hydrogel microspheres (CHMs) with removable superparamagnetic alginate coating (Alg-Mag-CHMs) to offer a minimal distance and easy separation. ^[8] Our in vitro 3D coculture data show that induction of DR5 in glioma cells with NEO214 and TRAIL cause tumor cell death very effectively and specifically for glioma cells. ^[9] The authors also demonstrate that upon immobilization of endothelial cells to the microwells, a second population of cells, namely, pancreatic islets could be introduced, thus producing a 3D coculture platform. ^[10]
